CFTR Deficiency Affects Glucose Homeostasis via Regulating GLUT4 Plasma Membrane Transportation

Cystic Fibrosis (CF) is an autosomal recessive disorder caused by mutations in the Transmembrane Conductance Regulator (CFTR) gene. CF-related diabetes (CFRD) one of most prevalent comorbidities CF. Altered glucose homeostasis has been reported CF patients. The mechanism not fully elucidated. Besides consequence pancreatic endocrine dysfunction, we focus on insulin-responsive tissues and transportation to explain alteration CFRD. Herein, found that CFTR knockout mice exhibited insulin resistance tolerance. Furthermore, demonstrated insulin-induced transporter 4 (GLUT4) translocation cell membrane was abnormal muscle fibers, suggesting defective intracellular GLUT4 may be cause impaired responses homeostasis. We further PI(4,5)P 2 could rescue related transportation, regulate cellular level through PIP5KA, a down-stream signal CFTR. Our results revealed new regulation, which helps
